Start From Nature
Quinine, extracted from the bark of the cinchona tree, has been used to treat malaria for over 300 years. Its quinoline ring system is the molecular scaffold responsible for disrupting the parasite's ability to detoxify heme — a critical mechanism of action.
Identify the Pharmacophore
Biomimetic analysis reveals that the quinoline core is the essential pharmacophore. Quinine's complex quinuclidine bicycle, while important for potency, can be replaced with simpler structural elements that maintain or improve the drug profile.
Engineer a Better Drug
Chloroquine retains the quinoline pharmacophore but replaces the complex bicyclic system with a simple aminoalkyl chain. A chlorine substituent enhances potency. The result: a fully synthetic drug that is cheaper to produce, orally bioavailable, and was the world's most widely used antimalarial for decades.
